اثر هارمان، نورهارمان و هارمین بر درد القاء شده با صفحه داغ و فرمالین در موش
Effects of Harmane, Norharman and Harmine on the Hot–Plate and Formalin-Induced Nociceptions in Mice

سابقه و هدف : هارمان، نورهارمان و هارمین اعضاء بتاکربولینی خانواده گیاه اسپند (پگانوم هارمالا، زیگوفیلاسه) میباشند. آلکالوئیدهای بتاکربولینی به عنوان آگونیستهای معکوس به جایگاه بنزودیازپینی گیرندههای گاما آمینوبوتیریک اسید نوع A (GABAA) متصل میشوند. این یافته، پیشنهاد میکند که هارمان، نورهارمان و هارمین میتوانند درد ناشی از صفحه داغ و فرمالین را در موشها کاهش دهند. در این مطالعه، اثر ضد دردی هارمان، نورهارمان و هارمین در موشها با استفاده از تست صفحه داغ و فرمالین مورد ارزیابی قرار گرفت.
مواد و روشها: همه آزمایشات بر روی موشهای نر از نژاد Balb/C (20 الی 25 گرم) انجام گرفت. اثر ضددردی داروها در تست صفحه داغ با استفاده از دستگاه صفحه داغ (هاروارد-انگلستان) مورد ارزیابی قرار گرفت. درجه حرارت صفحه داغ با ترموستات در دمای 5/0 ± 5/52 درجه سانتیگراد تنظیم گردید. زمان عکسالعمل لیسیدن یا لگد زدن پاهای جلو یا عقب حیوان در زمانهای مختلف پس از تزریق داروها ثبت گردید. برای جلوگیری از آسیب بافتی یک زمان چهل و پنج ثانیهای برای قطع آزمایش در نظر گرفته میشد. در تست فرمالین، کل زمان سپری شده برای لیسیدن پا در زمان های 0 و 5 دقیقه (بهعنوان فاز اولیه) و هر 5 دقیقه بین دقایق 15 تا 50 (بهعنوان فاز تأخیری) پس از تزریق فرمالین ثبت گردید. کاهش مدت زمان لیسیدن پا نشان دهنده پاسخ ضد دردی بود.
یافتهها: در تست صفحه داغ، تزریق داخل صفاقی هارمان (5 الی 20 میلیگرم/ کیلوگرم برای 7 موش در هر گروه)، نورهارمان (5 الی 15 میلیگرم/ کیلوگرم برای 7 موش در هر گروه) و هارمین (10 و 15 میلیگرم/ کیلوگرم برای 7 موش در هر گروه) بهطور معنیدار اثر ضد دردی اعمال نمود. اثر ضد دردی هارمان، نورهارمان و هارمین توسط فلومازنیل (2 میلیگرم/ کیلوگرم، داخل صفاقی) و نالوکسون (5 میلیگرم/ کیلوگرم، داخل صفاقی) آنتاگونیزه شد. در تست فرمالین، تزریق داخل صفاقی دوزهای 5/2، 5 و 10میلیگرم/ کیلوگرم هارمان، نورهارمان و هارمین بهطور معنیدار اثر ضددردی ایجاد کرد. اثر ضددردی هارمان، نورهارمان و هارمین توسط فلومازنیل (5 میلیگرم/ کیلوگرم، داخل صفاقی) آنتاگونیزه شد.
استنتاج: اثر ضد دردی هارمان، نورهارمان و هارمین ممکن است به صورت واسطه از طریق مکانیسم آگونیستی معکوس در سطح گیرندههای بنزودیازپینی انجام شود.
Background and purpose: Harmane, norharmane and harmine are -carboline members of the family of Harmalas alkaloids (Peganum harmala, Zygophillaceae). The -carboline alkaloids bind to benzodiazepine site of the γ-aminobutyric acid type A (GABAA) receptors as inverse agonists. This finding suggests that the harmane, norharmane and harmine might attenuate the hot-plate and formalin-induced nociceptions in mice. This study assessed the antinociceptive effects of harmane, norharmane and harmine in the mice used in hot-plate and formalin tests.
Materials and methods: The experiment was carried out on male BALB/C mice (20-25 g). In the hot-plate test, antinociceptive effects of drugs were assessed using a hot plate apparatus (Harvard, UK). The hot-plate temperature was thermostatically set at 52.5 ±5 °C. The latency to licking or kicking of the fore or hind paws was recorded at various times after drug injection. To avoid tissue damage a cut-off time of 45 seconds was imposed. In the formalin test, total time spent licking injected paw was recorded in 5 min intervals from 0-5 min (as early phase) and 15-50 min (as late phase) after injection of formalin. A decrease in the duration of the time spent licking indicated an antinociceptive response.
Results: In the hot-plate test i.p. injection of harmane (5-20 mg/kg, seven mice per group), norharmane (5-15 mg/kg, seven mice per group) and harmine (10 and 15 mg/kg, seven mice per group), led
to a significant antinociceptive effect. The antinociceptive effects of harmane, norharmane and harmine
were antagonized by flumazenil (2 mg/kg, i.p.) and naloxone (5mg/kg, i.p.). In the formalin test, i.p. injection of the doses of 2.5-5-10 mg/kg, harmane, norharmane and harmine resulted in a significant antinociceptive effect. The antinociceptive effects of harmane, norharmane and harmine were antagonized by flumazenil (5 mg/kg, i.p.).
Conclusion: The results suggest that the antinociceptive effects of harmane, norharmane and harmine may be mediated through an inverse agonistic mechanism located in the benzodiazepine receptors.
