fa اثرات ژل رویال بر درصد سلول‌های آپوپتوزی و بیان ژن‌های آپوپتوزی بافت بیضه در موش‌های نر نژاد Balb/c تیمار شده با داروی دانوروبیسین Effects of Royal Jelly on Apoptosis and the Expression of Apoptotic Genes on Testicular Tissue in Male Balb/C Mice Treated with Daunorubicin آسيب شناسي(پاتولوژی) Pathology پژوهشي-کامل Research(Original)  سابقه و هدف: دانوروبیسین یک داروی مؤثر در شیمی‌درمانی است، اما استفاده بالینی از آن به دلیل عوارض جانبی جدی، دارای محدودیت است. به‌نظر می‌رسد که ژل رویال؛ یکی از ترکیبات طبیعی حاصل از زنبور عسل، می‌تواند در جلوگیری از آسیب‌های ناشی از دانوروبیسین مفید باشد. بنابراین، مطالعه حاضر با هدف ارزیابی اثرات ژل رویال بر درصد سلول‌های آپوپتوزی و نیز میزان بیان ژن‌های دخیل در آپوپتوز در موش‌های تیمار شده با دانوروبیسین انجام شد. مواد و روش‌ها: در این مطالعه تجربی 77 سر موش نر نژاد Balb/c به طور تصادفی به 7 گروه (11 موش در هر گروه) شامل شم، کنترل، ژل رویال (50 و 100میلی‌گرم/کیلوگرم)، دانوروبیسین (2 میلی‌گرم/کیلوگرم) ، ژل رویال (50 و 100میلی‌گرم/کیلوگرم) + دانوروبیسین تقسیم شدند. موش‌ها به‌مدت ۸ هفته سالین ( 0/09 درصد) و دانوروبیسین را به‌صورت داخل صفاقی و ژل رویال را به‌شکل خوراکی دریافت نمودند. جهت تعیین درصد سلول‌های آپوپتوزی و بیان ژن‌های مرتبط با آپوپتوز در بیضه به‌ترتیب از روش الایزا و تکنیک Real-time PCR استفاده شد. تجزیه و تحلیل داده‌ها توسط آنالیز واریانس یک‌طرفه و تست توکی با نرم‌افزارSPSS انجام شد. یافته‌ها: دانوروبیسین سبب افزایش معنی‌دار درصد سلول‌های آپوپتوز و بیان ژن‌های p53، Bax، caspase 3، caspase 8 و caspase 9 (0/01>P) و کاهش معنی‌دار بیان ژن bcl-2 (0/01>P) شد، درحالی‌که در گروه درمان این تغییرات با تجویز ژل رویال تعدیل شد. استنتاج: تجویز ژل رویال با کاهش درصد سلول‌های آپوپتوزی و اثر بر بیان ژن‌های دخیل در آپوپتوز، سبب بهبود آسیب‌های ناشی از دانوروبیسین روی بافت بیضه موش‌ می‌شود.    Background and purpose: Daunorubicin is an effective drug in chemotherapy, but its clinical use is limited due to serious side effects. Royal jelly as a natural compound obtained from bees can be helpful in preventing the damage caused by daunorubicin. The present study aimed to evaluate the effects of royal jelly on the number of apoptotic cells and the expression of genes involved in apoptosis in mice treated with daunorubicin. Materials and methods: In this experimental study, 77 Balb/c mice were randomly divided into seven groups (n= 11 per group): sham, control, royal jelly (50 and 100 mg/kg), daunorubicin (2 mg/kg), and royal jelly (50 and 100 mg/kg) + daunorubicin. The mice received saline (0.09%), IP daunorubicin, and royal jelly orally for eight weeks. To estimate the number of apoptotic cells and the expression of genes related to apoptosis in testis, we used ELISA test and Real-time PCR assay, respectively. Data analysis was done applying One-way Analysis of Variance and Tukey's test in SPSS. Results: Daunorubicin caused significant increase in the number of apoptotic cells, and expression levels of p53, Bax, caspase 3, caspase 8, and caspase 9 (P<0.01) and a significant decrease in the bcl-2 gene expression (P<0.01).  In treatment groups, these changes were moderated by royal jelly.
