fa مقایسه تأثیر دهانشویه گیاهی حاوی عصاره هیدروالکلی سعدکوفی و آویشن با کلرهگزیدین بر پاتوژن‌های شایع پریودنتال Comparing the Effect of Herbal Mouthwash Containing Hydroalcoholic Extracts of Cyperus rotundus L. and Thymus vulgaris L. and Chlorhexidine Mouthwash on Common Periodontal Pathogens داروسازی Pharmacy پژوهشي-کامل Research(Original) <div>سابقه و هدف: ژنژیویت بیماری التهاب لثه است که ناشی از تجمع پلاک‌های دندانی می‌باشد. در صورتی که پلاک و جرم به زیر خط لثه گسترش پیدا کند، می‌تواند به بیماری مزمن پریودنتال تبدیل شود. شایع‌ترین میکروب‌هایی که در ایجاد بیماری پریودنتال نقش دارند، شامل Streptococcus mutans، Streptococcus pyogenes و Candida albicans است. با توجه به عوارض درمان رایج با دهانشویه کلرهگزیدین، به منظور مبارزه با این میکروب‌ها، دستیابی به درمان‌های جایگزین، امری مطلوب می‌باشد. لذا این مطالعه به مقایسه اثر ضدمیکروبی دهانشویه گیاهی حاوی عصاره هیدروالکلی سعدکوفی و آویشن با دهانشویه کلرهگزیدین بر پاتوژن‌های شایع پریودنتال پرداخت. مواد و روش‌ها: ابتدا گیاهان سعدکوفی و آویشن جمع‌آوری و خرد شدند و عصاره هیدروالکلی مخلوط دو گیاه و عصاره متانولی هرگیاه، به صورت جداگانه، گرفته شد. سپس با استفاده از اکسپیان‌های مختلف، محلول دهانشویه ترکیبی ساخته شد. برای ارزیابی اثر ضد میکروب، از میکروارگانیسم‌های S. mutans، S. pyogenes و C. albicans استفاده و پس از تعیین MIC/MBC، اثر دهانشویه بر تشکیل و تخریب بیوفیلم S. mutans بررسی شد. یافته‌ها: مقادیر MIC دهانشویه علیه S. mutans، S. pyogenes و C. albicans به ترتیب معادل 625 /0،  039 /0 و 2/5 mg/mL حاصل شد. هم‌چنین، میزان MBC دهانشویه برای هر یک از میکروارگانیسم‌ها به ترتیب 1/25، 078 /0 و ۱۰mg/mL به‌دست آمد. از سوی دیگر، تشکیل بیوفیلم توسط فرمولاسیون دهانشویه به‌طور معناداری کم‌تر از کنترل مثبت گردید(0/05>P) و هیچ تفاوت معناداری با کنترل منفی مشاهده نشد(0/05<P). اما در مورد تخریب بیوفیلم از لحاظ آماری تفاوت معناداری بین گروه دهانشویه و کنترل مثبت وجود نداشت(0/05<P). استنتاج: براساس نتایج این مطالعه، دهانشویه گیاهی مورد استفاده می‌تواند به‌صورت قابل توجهی اثرات ضدمیکروب و ضد بیوفیلم داشته باشد، به گونه‌ای که در پیشگیری از تشکیل بیوفیلم حتی نسبت به درمان استاندارد کلرهگزیدین، بسیار مؤثرتر عمل کرد.  </div>  Background and purpose: Gingivitis is an inflammatory disease of the gums caused by accumulation of dental plaque. It can become a chronic periodontal disease if plaque spread below the gum line. The most common pathogens that contribute to periodontal disease include Streptococcus mutans, Streptococcus pyogenes, and Candida albicans. Due to the side effects of common treatment by chlorhexidine mouthwash, novel treatments to fight such microbes are much of interest. This study aimed at comparing the antimicrobial effect of herbal mouthwash containing the extracts of nut-grass and thyme and chlorhexidine mouthwash on common periodontal pathogens. Materials and methods: First, Cyperus rotundus and Thymus vulgaris were collected, powdered, and the hydro alcoholic extract of the mixture of two plants and the methanol extract of each plant were taken. Then, a mouthwash containing combined extract of the two plants was prepared using different exipients. To evaluate the antimicrobial effect of the extracts, S. mutans, S. pyogenes, and C. albicans were used. After determining the minimum inhibitory concentration (MIC) and minimum bactericidal concentration (MBC), the effect of mouthwash on S. mutans biofilm formation and degradation was investigated. Results: The MIC of the mouthwash formulation against S. mutans, S. pyogenes, and C. albicans were 0.625, 0.039, and 2.5 mg/ml, respectively. The MBC of the mouthwash for each microorganism was 1.25, 0.078, and 10 mg/ml, respectively. The herbal mouthwash significantly inhibited biofilm formation compared to the positive control (P<0.05) which showed no significant difference compared to the negative control (P>0.05). Findings showed no significant difference in biofilm destruction between the herbal mouthwash and the positive control (P>0.05).
