fa شناسایی جهش های موجود در ژن نورآمینیداز ویروس‌های آنفلوانزا A/H3N2 در نمونه های استان مازندران طی سال های 1395 لغایت 1397 Identification of Neuraminidase Gene Mutations of Influenza A/H3N2 Isolates in Mazandaran Province, 2016-2018 ويروس شناسي پزشکي Medical Virology پژوهشي-کامل Research(Original) سابقه و هدف: بیماری آنفلوانزا یکی از عفونت‌های ویروسی دستگاه تنفسی است که هر ساله باعث مرگ در گروه‌های پر خطر می‌شود. تغییرات ژنومی ویروس آنفلوانزا آن را مستعد مقاومت دارویی می‌کند، بنابراین نظارت مداوم به منظور آگاهی از اثربخشی داروهای موجود بسیار احساس می‌شود. در مطالعه حاضر شناسایی جهش‌های موجود در ویروس آنفلوآنزای A/H3N2 در نمونه‌های استان مازندران از سال 1395 لغایت 1397 مورد بررسی قرار گرفته است. مواد و روش‌ها: از بین نمونه‌های بیماران مبتلا به آنفلوانزا بستری در بیمارستان‌های استان مازندران، 20 نمونه تایید شده H3N2 وارد مطالعه شدند. با استفاده از پرایمرهای طراحی شده، تست‌های PCR و الکتروفورز در ناحیه‌ای از ژن نورامینیداز که حاوی موتاسیون‌های مورد نظر بوده است، انجام شد. سپس محصول PCR جهت مشخص شدن موتاسیون‌ها تعیین سکانس شد. یافته‌ها: در مجموع 57 جهش در سطح نوکلئوتیدها شناسایی گردید که در این میان 19 جهش در تمام ایزوله‌ها، 13 جهش در اکثر ایزوله‌ها و 25 جهش بصورت انفرادی در برخی از ایزوله‌ها مشاهده گردید. در مجموع 28 جهش در سطح اسیدآمینه شناسایی گردید که در این میان 13 جهش در تمام ایزوله‌ها، 6 جهش در اکثر ایزوله‌ها و 9 جهش به‌صورت انفرادی در برخی از ایزوله‌ها مشاهده گردید. استنتاج: با وجود حفاظت شده بودن توالی ناحیه اکتیو سایت در ساب تایپ‌های مختلف از ژن نورامینیداز این ناحیه همچنان دستخوش موتاسیون و تغییرات ژنومی قرار دارد که نشان دهنده تغییرات در ناحیه اکتیو سایت ژن نورامینیداز می‌باشد.    Background and purpose: Influenza is one of the viral infections of the respiratory system, which causes death in high-risk groups every year. The genomic changes of influenza virus make it susceptible to drug resistance, therefore, continuous monitoring of the influenza virus is highly important in order to recognize the efficacy of available drugs. The current study investigated the neuraminidase gene mutations of Influenza A/H3N2 in Mazandaran province between 2016 and 2018. Materials and methods: Twenty Influenza A/H3N2 samples of patients hospitalized in Mazandaran province were included in the study. PCR and electrophoresis tests were performed using primers designed for the region of the neuraminidase gene that contained the desired mutations. Then, the PCR product was sequenced to determine the mutations. Results: A total of 57 mutations were detected at the level of nucleotides, among which 19 were observed in all isolates, 13 were seen in most isolates, and 25 single mutations that were identified in some isolates. A total of 28 mutations were detected at the amino acid level, among which 13 mutations were observed in all isolates, 6 were detected in most isolates, and 9 single mutations seen in some isolates.
