fa بررسی بیان ژن های lncRNA HOTAIR و lncRNA PVT1 در افراد مبتلا به نفروپاتی دیابتی Investigation of the Expression of lncRNA HOTAIR and lncRNA PVT1 in Individuals with Diabetic Nephropathy ژنتیک genetic پژوهشي-کامل Research(Original) سابقه و هدف: نفروپاتی دیابتی یکی از عوارض میکروواسکولار دیابت نوع دو است که می‌تواند منجر به نارسایی کلیوی گردد. در این مطالعه به بررسی بیان ژن های lncRNA HOTAIR و lncRNA PVT1 در افراد مبتلا به DN (دیابت نوع دو با نفروپاتی دیابتی)، افراد مبتلا به T2DM (دیابت نوع دو بدون نفروپاتی دیابتی )، گروه Pre-diabetes (پره دیابتی) و گروه Control (افراد سالم) پرداخته شد. علاوه بر این، میزان بیانlncRNA HOTAIR  و lncRNA PVT1 در سلول‌های HEK-293 تحت تیمار با گلوکز بالا و گلوکز طبیعی ارزیابی شد. مواد و روش ‌ها: در این مطالعه مقطعی، سطوح بیان ژن‌های lncRNA HOTAIR و lncRNA PVT1 در بین چهار گروه، 50 فرد سالم، 50 فرد مبتلا به Pre-diabetes، 50 بیمار مبتلا به T2DM و 50 بیمار مبتلا به DN، مقایسه گردید. هم‌چنین برای القای مدل DN در محیط آزمایشگاهی، سلول‌های HEK-293 به میزان 72 ساعت در شرایط گلوکز بالا (100mmolar/l) و گلوکز طبیعی تیمار شدند، و میزان بیان ژن‌های هدف در سلول‌های مورد نظر اندازه‌گیری شد. یافته‌ها: میزان بیان lncRNA PVT1 به میزان قابل ملاحظه‌ای در افراد مبتلا به DN نسبت به گروه سالم و pre-diabetes افزایش می یابد ولی تفاوت معنی‌داری با گروه T2DM نشان نمی دهد. هم‌چنین میزان بیان lncRNA HOTAIR در گروه DN در مقایسه با سایر گروه‌ها افزایش یافت. هم‌چنین، گلوکز بالا باعث افزایش میزان بیان lncRNA PVT1 شد، در حالی که تغییری در میزان بیان lncRNA HOTAIR ایجاد نکرد. استنتاج: ژن‌های lncRNA HOTAIR و lncRNA PVT1 در پاتوژنز DN دخالت دارند، و هم‌چنین lncRNA PVT1 نسبت به lncRNA HOTAIR همراهی قوی‌تری با تغییرات سلولی ناشی از گلوکز دارد.   Background and purpose: Diabetic nephropathy (DN) is a major microvascular complication of type 2 diabetes mellitus (T2DM) that can lead to kidney failure. In this study, the expressions of the lncRNA HOTAIR and PVT1 genes were investigated in individuals with type 2 diabetes with DN, individuals with type 2 diabetes without DN, pre-diabetic individuals, and healthy controls. The study also examined the expression levels of lncRNA HOTAIR and PVT1 in HEK-293 cells treated with high glucose (HG) and normal glucose (NG). Materials and methods: In this cross-sectional study, the expression levels of the lncRNA HOTAIR and PVT1 genes were compared among four groups: 50 healthy individuals, 50 individuals with prediabetes, 50 patients with T2DM, and 50 patients with DN. To model DN in vitro, HEK-293 cells were cultured under high-glucose (HG; 100 mmol/L) and normal-glucose (NG) conditions for 72 hours, and the expression levels of HOTAIR and PVT1 were measured in the treated cells. Results: The current work showed that the expression level of lncRNA PVT1 was significantly increased in individuals with DN compared with the healthy and prediabetic groups, but there was no significant difference when compared with the T2DM group. The analysis also indicated that the expression level of lncRNA HOTAIR was elevated in the DN group compared with the other groups. Under high-glucose (HG) conditions, an increase in lncRNA PVT1 expression was observed, while no change was detected in the expression level of lncRNA HOTAIR.
