fa شناسایی lncRNA LINC02747 به عنوان یک نشانگر زیستی جدید بالقوه برای سرطان پستان Identification of lncRNA LINC02747 as a Novel Potential Biomarker for Breast Cancer بيولوژي Biology پژوهشي-کامل Research(Original) سابقه و هدف: سرطان پستان علت اصلی مرگ ناشی از سرطان در میان زنان جهان است. در نتیجه، شناسایی زود هنگام سرطان پستان برای افزایش بقای بیمار مهم است و نشانگرها به عنوان ابزارهای امیدوارکننده برای دستیابی به این هدف می‌باشند. این مطالعه با هدف شناساییlncRNA ‌های مرتبط با سرطان پستان به عنوان نشانگرهای پیش‌آگهی جدید برای این بیماری انجام پذیرفت، زیرا نقش مهمی در پاتولوژی سرطان دارند. مواد و روش ‌ها: در این مطالعه تجربی– آزمایشگاهی، بررسی داده‌های بیان ژن از پایگاه داده TCGA آنالیز گردید، mRNA های با بیان متفاوت، miRNAها و lncRNAها را شناسایی و یک شبکه  RNA های درون‌زا رقیب (ceRNA) برای شناسایی نشانگرهای بالقوه مرتبط با تشخیص و پیش آگهی سرطان پستان ایجاد شد. بیان lncRNA هدف در سلول های 7 MCF و HEK293T برای اعتبارسنجی داده‌های بیوانفورماتیکی بررسی شد. یافته‌ها: بررسی داده‌های TCGA نشان داد بیانLINC02747، که قوی‌ترین تعامل را با سایر miRNAها و mRNAهای شناسایی ‌شده توسط شبکه ceRNA داشت، در سرطان پستان افزایش یافته است. هم‌چنین، طبق شبکه ceRNA، hsa-miR-195- 5p و ده mRNA شامل CCNB1، CCNB2، CCNA2، PLK1، CDC6، CDC20، CDC45، BUB1B، CDT1وESPL1  باlncRNA  فوق ‌الذکر مرتبط بودند، که با کاهش بیان microRNA و افزایش بیان mRNA همراه بود. آنالیز RT-qPCR یافته‌‌های بیوانفورماتیک مطالعه حاضر را تأیید کرد. آنالیز ROC نشان داد که LINC02747 یک نشانگر امیدوار کننده برای سرطان پستان است. استنتاج: بیان بالای LINC02747 ممکن است به عنوان یک نشانگر مولکولی برای سرطان پستان عمل کند، شاید از طریق اسفنج hsa-miR-195-5p، بیان mRNAهای هدف را که در فرآیند سرطان‌زایی مؤثر هستند، افزایش می‌دهد.   Background and purpose: Breast cancer (BC) is the leading cause of cancer-related mortality among women worldwide. Consequently, the early identification of breast cancer is crucial for improving patient survival, and biomarkers represent promising tools for achieving this objective. This study aims to identify long non-coding RNAs (lncRNAs) associated with breast cancer as potential prognostic biomarkers, given their significant role in cancer pathogenesis. Materials and methods: This experimental study analyzed gene expression data from the TCGA database, identified differentially expressed mRNAs, miRNAs, and lncRNAs, and constructed a competing endogenous RNA (ceRNA) network to identify potential biomarkers associated with the diagnosis and prognosis of breast cancer. The expression of the target lncRNA in MCF-7 and HEK293T cell lines was examined to validate the bioinformatics findings. Results: Analysis of TCGA data revealed that LINC02747 expression, which showed the strongest interactions with other miRNAs and mRNAs identified within the ceRNA network, was upregulated in breast cancer. Furthermore, according to the ceRNA network, hsa-miR-195-5p and ten mRNAs- CCNB1, CCNB2, CCNA2, PLK1, CDC6, CDC20, CDC45, BUB1B, CDT1, and ESPL1- were linked to the aforementioned lncRNA, with decreased microRNA expression and increased mRNA expression. The RT-qPCR analysis validated these bioinformatics findings. ROC analysis indicated that LINC02747 is a highly promising biomarker for breast cancer.
