fa بررسی اثر محافظتی آلفا لیپوئیک اسید بر آسیب کبدی ناشی از بیسفنل-آ از طریق تعدیل مسیر Nrf2 و شاخص‌های استرس اکسیداتیو در مدل موش کوچک آزمایشگاهی The Hepatoprotective Effect of Alpha-Lipoic Acid against Bisphenol A-Induced Liver Injury in Mice: Modulation of the Nrf2 Pathway and Oxidative Stress سم شناسی toxicology پژوهشي-کامل Research(Original) سابقه و هدف: بیسفنل آ (BPA) به عنوان یک ترکیب مختل‌ کننده غدد درون‌ریز، از طریق القای استرس اکسیداتیو و التهاب باعث سمیت کبدی می‌شود. آلفا لیپوئیک اسید (α-LA) ترکیبی با خاصیت آنتی اکسیدانی است که می تواند از آسیب کبدی ناشی از عوامل آسیب زا جلوگیری کند. این مطالعه با هدف بررسی نقش محافظتی α-LA در برابر آسیب کبدی ناشی از BPA در موش کوچک آزمایشگاهی، انجام پذیرفت مواد و روش ‌ها: در این مطالعه تجربی، 28 سر موش نژاد NMRI به صورت تصادفی به چهار گروه کنترل، دریافت‌ کننده (mg/kg 100) α-LA، دریافت‌کننده (mg/kg 300) BPA و درمان ترکیبی (BPA+α-LA) تقسیم شدند. پس از 35 روز، نمونه‌های خون و بافت کبد جمع‌آوری شد. سطوح سرمی AST و ALT، شاخص‌های استرس اکسیداتیو MDA، SOD و CAT و بیان ژن‌های Nrf2 و NF-κB با روشRT-PCR  (واکنش زنجیره‌ای پلیمراز معکوس) اندازه‌گیری شد. آسیب بافتی با رنگ‌آمیزی  H&Eارزیابی شد. داده‌ها با نرم‌افزار SPSS و آزمون‌هایANOVA  یک‌طرفه و Tukey's تحلیل شدند. یافته‌ها: تجویز BPA به طور معنی‌داری باعث افزایش آنزیم‌های کبدی، سطح MDA و بیان NF-κB، و نیز کاهش فعالیت آنزیم‌های آنتی‌اکسیدانی و بیان Nrf2 گردید. هم‌زمان، آسیب هیستوپاتولوژیک واضحی شامل نفوذ التهابی و احتقان عروقی مشاهده شد. درمان با α-LA به طور قابل توجهی این اختلالات را معکوس کرد و موجب کاهش نشانگرهای آسیب کبدی، بهبود شاخص‌های استرس اکسیداتیو، افزایش بیان Nrf2 و کاهش بیان NF-κB شد. استنتاج: α-LA از طریق تقویت سیستم دفاع آنتی‌اکسیدانی (احتمالاً با فعال‌سازی مسیر Nrf2 )، مهار پراکسیداسیون لیپیدی و تعدیل پاسخ التهابی (با مهار مسیر NF-κB)، اثر محافظتی قابل توجهی در برابر سمیت کبدی ناشی از BPA دارد. یافته‌ها از کاربرد α-LA به عنوان یک راهبرد درمانی امیدوار کننده برای پیشگیری از مواد شیمیایی حمایت می‌کنند. این ترکیب می‌تواند به عنوان یک عامل درمانی بالقوه در برابر آسیب‌های ناشی از مواد مختل‌کننده غدد درون‌ریز مورد توجه قرار گیرد. Background and purpose: Bisphenol A (BPA), an endocrine-disrupting compound, causes hepatotoxicity through oxidative stress and inflammation. Alpha-lipoic acid (α-LA) is a potent antioxidant shown to protect against liver injury induced by various toxic agents. This study aimed to evaluate the protective effect of α-LA against BPA-induced liver damage in mice. Materials and methods: In this experimental study, 28 NMRI mice were randomly assigned to four groups: control, α-LA (100 mg/kg), BPA (300 mg/kg), and BPA + α-LA co-treatment. After 35 days, blood and liver tissue samples were collected. Serum activities of AST and ALT, oxidative stress markers (MDA, SOD, and CAT), and the expression of the Nrf2 and NF-κB genes were evaluated using RT-PCR. Histopathological changes were evaluated by H&E staining. Data were analyzed using SPSS software by one-way ANOVA followed by Tukey's post hoc test. Results: BPA administration significantly increased liver enzyme activities, MDA levels, and NF-κB expression, while decreasing antioxidant enzyme activities and Nrf2 expression. In addition, marked histopathological alterations, including inflammatory cell infiltration and vascular congestion, were observed. Co-treatment with α-LA significantly attenuated these disturbances by reducing hepatic injury markers, improving oxidative stress parameters, upregulating Nrf2 expression, and downregulating NF-κB expression. Conclusion: The findings indicate that α-LA exerts a significant protective effect against BPA-induced hepatotoxicity, possibly by enhancing the antioxidant defence system through activation of the Nrf2 pathway, inhibiting lipid peroxidation, and suppressing the inflammatory response via downregulation of the NF-κB pathway. These findings support the potential of α-LA as a promising therapeutic strategy for preventing chemically induced liver injury.
