fa بررسی مقایسه‌ای بروز GLUT1 در آملوبلاستومای توپر و یونی‌سیستیک آملوبلاستومای مورال و غیرمورال به روش ایمنوهیستوشیمی Comparative Study of GLUT1 Expression in Solid Ameloblastoma and Mural and Non-Mural Unicystic Ameloblastoma: An Immunohistochemical Study ﭘﺎﺗﻮﻟﻮژي دﻫﺎن و ﻓﻚ و ﺻﻮرت Oral and maxillofacial pathology پژوهشي-کامل Research(Original) سابقه و هدف: آملوبلاستومای توپر و یونی سیستیک آملوبلاستوما یکی از شایع‌ترین ضایعات در ناحیه فک می‌باشند. ورود گلوکز به سلول توسط گروهی از پروتئین‌های حامل غشایی به نام انتقال دهنده‌های گلوکز (GLUTs ) انجام می‌شود. بیان این حامل می‌تواند در ضایعات خوش‌خیم و به‌ویژه بدخیم به‌دلیل نیاز بیش‌تر به گلوکز افزایش یابد، در نتیجه این حامل به تامین انرژی لازم برای تومور کمک می‌کند. طبق مطالعات اخیر، میزان بیان GLUT1 در تشخیص ضایعات کمک کننده می باشد. این مطالعه با هدف بررسی مقایسه‌ای بیان GLUT1 در آملوبلاستومای توپر و انواع یونی‌سیستیک آملوبلاستوما به روش IHC، انجام پذیرفت. مواد و روش ‌ها: در این مطالعه‌ی توصیفی– تحلیلی از نوع مقطعی، 20 نمونه&lrm; آملوبلاستومای توپر و 20 نمونه&lrm; یونی‌سیستیک آملوبلاستوما (10 نمونه مورال و 10 نمونه غیرمورال) از آرشیو بخش آسیب شناسی دهان دانشکده دندانپزشکی دانشگاه علوم پزشکی اصفهان که به روش بیوپسی اکسیژنال تهیه شده بودند، انتخاب شده و با تکنیک ایمونوهیستوشیمی برای مارکر GLUT1 رنگ‌آمیزی و میزان بیان آن در سلول های اپیتلیالی ادنتوژنیک بر اساس شاخص SID توسط دو نفر پاتولوژیست دهان به‌طور همزمان بررسی شد. اطلاعات حاصل توسط آزمون‌های تی مستقل، کای دو و آزمون دقیق فیشر مورد بررسی آماری قرار گرفت و میزان 0/05P< معنی‌دار تلقی شد. یافته‌ها: در مطالعه حاضر، فراوانی سلول&rlm;های رنگ گرفته برای مارکر GLUT1 (0/001 P<)، شدت رنگ&lrm;پذیری (0/004=P ) و میانگین شاخص SID (0/001 P< ) در گروه آملوبلاستومای توپر به&lrm;طور معنی&rlm;داری بیش‌تر از انواع یونی‌سیستیک آملوبلاستوما بود. هم‌چنین میانگین شاخص SID براساس سن، جنسیت و مکان ضایعه تفاوت آماری معنی&lrm;داری نشان نداد (0/05<P). استنتاج: مطالعه حاضر افزایش بیان معنی‌دار GLUT1 را در آملوبلاستومای توپر نسبت به انواع یونی سیستیک آملوبلاستوما نشان داد. این افزایش بیان نشان می&lrm;دهد که احتمالا متابولیسم تغییر یافته گلوکز ممکن است یکی از عوامل مرتبط با رفتار بالینی آملوبلاستومای توپر باشد. Background and purpose: Ameloblastoma is one of the most common lesions of the jaw. Glucose entry into cells is mediated by a group of membrane-bound proteins called glucose transporters (GLUTs). The expression of these transporters may be increased in benign and, particularly, malignant lesions due to their increased demand for glucose; thus, GLUTs help provide the energy required for tumour growth and metabolism. According to recent studies, the expression level of GLUT1 may be useful in the diagnosis and evaluation of lesions. This study aimed to compare GLUT1 expression in solid and unicystic ameloblastoma types using immunohistochemistry (IHC). Materials and methods: In this descriptive-analytical cross-sectional study, 20 samples of solid ameloblastoma and 20 samples of unicystic ameloblastoma (10 mural and 10 non-mural) were selected from the archives of the Oral Pathology Department at Isfahan Dental School. The samples, which had been prepared using excisional biopsy, were stained using the IHC method for GLUT1 expression. The expression level of GLUT1 in odontogenic epithelial cells was evaluated based on the SID index by two oral pathologists. The obtained data were statistically analysed using independent t-tests, chi-square tests, the Mann-Whitney U test, and Fisher's exact test. A P-value of <0.05 was considered statistically significant. Results: The frequency of GLUT1-positive cells (P < 0.001), staining intensity (P = 0.004), and SID index (P < 0.001) in solid ameloblastoma samples was significantly higher than in unicystic ameloblastoma samples. Furthermore, the mean SID index did not show statistically significant differences according to age, sex, or lesion location (P > 0.05). Conclusion: The present study demonstrated a significant increase in GLUT1 expression in solid ameloblastoma compared with unicystic ameloblastoma.
