fa بررسی اثرات ضدالتهابی و ضد دردی عصاره گیاه گلوسیوم گرندیفلوروم (Glaucium grandiflorum) پژوهشي-کامل Research(Original) <div style="text-align: justify direction: rtl"> <span style="font-weight: bold">سابقه و هدف :</span>گونه های مختلف گیاه گلوسیوم در طب سنتی ایران با کاربردهای مختلف از جمله در درمان درماتیت استفاده می شوند. با توجه به خواص عصاره های گیاهان خانواده خشخاش، در این پژوهش به بررسی اثرات ضد التهابی و ضد دردی عصاره گیاه گلوسیوم گرندیفلوروم پرداخته شده است.<br><span style="font-weight: bold">مواد و روش ها :</span> عصاره گیری با استفاده از متانول و با روش پرکولاسیون انجام پذیرفت. جهت بررسی اثرات ضد التهابی عصاره با آزمون کاراژینال، دوزهای 4، 10، 20، 40، 80، 160 و 200 میلی گرم بر کیلوگرم وزن بدن عصاره در مقایسه با دوزهای 2، 4، 6، 8 و 10 میلی گرم بر کیلوگرم وزن بدن ایندومتاسین مورد مطالعه قرار گرفت. اثر ضد دردی عصاره با آزمون فرمالین و صفحه داغ(Hot plate) در دوزهای 60، 70، 80 و 90 میلی گرم بر کیلوگرم وزن بدن عصاره در مقایسه با دوزهای 5 و 10 میلی گرم بر کیلوگرم وزن بدن مورفین مورد مطالعه قرار گرفت. آزمون روتارود (Rotarod) جهت بررسی هماهنگی حرکتی مورد استفاده قرار گرفت. سمیت حاد(LD50) عصاره با روش پروبیت تعیین گردید. جهت آنالیز داده ها از آنالیز واریانس و آزمون Newman- Keuls با ارزش p<0.05 استفاده شد.<br><span style="font-weight: bold">یافته ها:</span> دوز 200 kg/mg عصاره اثرات ضدالتهاب بیشتری نسبت به دوزهای(p<0.01) 10 mg/kg و8 mg/kg و(p<0.001) ایندومتاسین نشان داد. تمامی دوزهای مورد استفاده اثرات ضددرد موثری نسبت به گروه کنترل نشان دادند(p<0.001) . اثرات ضد درد دوزهای 80 و 90 میلی گرم بر کیلوگرم وزن بدن عصاره در فاز اولیه بیشتر از دوز 5 kg/mg مرفین بود(p<0.001) . در فاز ثانویه اثرات ضد دردی دوزهای 70، 80 و 90 میلی گرم بر کیلوگرم وزن بدن بیشتر از اثر ضد درد دوز 5 kg/mg مرفین بود (p<0.001). بین دوزهای 80 و 90 میلی گرم بر کیلوگرم وزن بدن اختلاف معنی دار وجود نداشت. تجویز عصاره در دوزهای 60، 70، 80 و 90 میلی گرم بر کیلوگرم وزن بدن آستانه درد را در مقایسه با گروه کنترل در زمان 45 دقیقه افزایش داد. عصاره گیاه فوق اثری بر تغییر هماهنگی حرکات حیوان نداشت. سمیت حاد عصاره معادل797.94 kg/mg به صورت داخل صفاقی تعیین گردید.<br><span style="font-weight: bold">استنتاج: </span>نتایج حاصل می تواند گیاه فوق را با اثرات ضد التهاب و ضد درد معرفی نماید. </div> <span style="font-weight: bold">Background and purpose:</span> Various species of the plant grandiflorum have different applications in Ïran traditional medicine, such as in treatment of dermatitis. Çonsidering the therapeutic property of papuveraceae, this study was designed in order to determine the anti inflammatory and antinociceptive property of Glaucium grandiflorum. <br> <span style="font-weight: bold">Materials and methods :</span> Ëxtraction was done with methanol by percolation method. Ânti inflammatory property of the extract was determined by carrageenan test for the doses of 4,10,20, 40, 80, 160 and 200 mg/kg of body weight as compare to indometacin for the doses of 2. 4, 6, 8 and 90 mg/kg of body weight. Ântinociceptive property of the extract with formalin and hot plate was studied for the doses of 60,70,80 and 90 mg/kg of body weight as compare to morphine for the doses of 5 and 10 mg/kg of body weight. The integrity of motor coordination was assessed with a rotarod apparatus. Âcute toxicity (LD50) of the extract was determined by probit method. Data were analyzed by variance test and Newman- keuls test with P<0.05. <br> <span style="font-weight: bold">Results: </span>200 mg/kg dose of the extract had more anti inflammatory effect as compare to indomethacin at the doses of 10 mg/kg (P<0.01) and 8 mg/kg (P<0.001). Âll the used doses had more antinociceptive effect as compare to the control group, (P<0.001). Ântinociceptive effects of 80 and 90 mg/kg of body weight of the extract in the first phase was more than morphine for the dose of 5 mg/kg body weight. Ïn the second phase, the antinociceptive effect of 70, 80 and 90 mg/kg of body weight was more than morphine for the dose of 5 mg/kg of body weight P<0.001. There was no significant difference in the doses of 80 and 90 mg/kg of body weight. Âdministration of the extract with the doses of 60, 70,80 and 90 mg/kg of body weight, raised the pain threshold at 45 minutes of observation, as compare to the control group. Ëxtract fluid of this plant had no effect on motor coordination of the animals.
