اثر حفاظتی روی و سلنیوم در جلوگیری از سمیت کلیوی ناشی از سدیم والپروات با مکانیسم مهار استرس اکسیداتیو

<p dir="RTL">سابقه و هدف: سدیم والپروات یک داروی پر مصرف و مهم در درمان صرع و کنترل انواع خاصی از تشنج ها است. هدف از این مطالعه ارزیابی اثر حفاظتی روی و سلنیوم در سمیت کلیوی ناشی از سدیم والپروات در رت های ویستار میباشد.</p>
<p dir="RTL">مواد و روشها: در این مطالعه رتها به پنج گروه کنترل، سدیم والپروات (<span dir="LTR">mg/kg IP</span>200)، سدیم والپروات+ روی <span dir="LTR">mg/kgIP</span>10، سدیم والپروات+ سلنیوم <span dir="LTR">mg/kg IP</span>1، سدیم والپروات+روی+سلنیوم تقسیم شدند. در پایان بعد از دریافت والپروات 4 هفته متوالی، حیوانات کشته شده و بافت کلیه جدا شدو پارامترهای استرس اکسیداتیو شامل اندازهگیری محتوای گلوتاتیون، پراکسیداسیون لیپیدیوپروتئین کربونیل و خون جهت پارامترهای بیوشیمیایی (اوره و کراتینین) مورد بررسی قرار گرفت.</p>
<p dir="RTL">یافتهها: دریافت سدیم والپروات برای 4 هفته متوالی منجر به افزایش مارکرهای کلیوی اوره و کراتینین شد. همچنین استرس اکسیداتیو درگروه والپروات با افزایش پراکسیداسیون لیپیدی، پروتئین کربونیل و اکسیداسیون گلوتاتیون مشهود است. دریافت روی و سلنیوم قادر به حفاظت علیه بدتر شدن در مارکرهای کلیوی و مهار افزایش مارکرهای استرس اکسیداتیو شد.</p>
<p dir="RTL">استنتاج: مطالعه ما نشان داد که تخریب اکسیداتیو نقش حیاتی در سمیت کلیوی ناشی از والپروات دارد. که بهطور چشمگیری با دریافت روی و سلنیوم مهار میشود. بنابراین مکملهای روی و سلنیوم میتواند برای حفاظت از سمیت کلیوی ناشی از والپروات پیشنهاد شود.</p>
<p><strong>Background and purpose:</strong> Valproic acid (VPA) is used worldwide as a major drug in the intervention of epilepsy and in control of several kinds of seizures. The aim of our investigation was to evaluate the nephrotoxic potential of VPA and protective effects of zinc and selenium against VPA-induced nephrotoxicity in Wistar rats.</p>
<p><strong>Materials and methods:</strong> In this study, the animals were divided into five groups: control, VPA (200 mg/kg IP), VPA + Zn (10 mg/kg IP), PA + Se (1 mg/kg IP), and VPA + Zn + Se. After the administration of VPA for 4 consecutive weeks, the animals were killed and kidney tissues were separated. Finally, oxidative stress markers including glutathione (GSH) content, lipid peroxidation (LPO), protein carbonyl (PCO) were measured and blood was taken for measuring biochemical markers (BUN and Cr).</p>
<p><strong>Results:</strong> The administration of VPA for 4 consecutive weeks resulted in an increase in kidney marker (BUN and Cr). Also, oxidative stress was evident in VPA group by increased lipid peroxidation (LPO), protein carbonyl (PCO) and glutathione (GSH) oxidation. Zn and Se administration was able to protect against deterioration in kidney markers and suppressed the increase in oxidative stress markers.</p>
<p><strong>Conclusion:</strong> Our study showed the critical role of oxidative damage in Valproate-induced nephrotoxicity that markedly inhibited by administration of Zn and Se. Therefore, Zn and Se supplementation could be suggested for prevention of valproate-induced nephrotoxicity.</p>
