Background and purpose: Toxoplasmosis, caused by the intracellular protozoan parasite Toxoplasma gondii, is one of the most prevalent parasitic infections worldwide. The limitations of currently available therapeutic agents, including toxicity, drug resistance, and high treatment costs, highlight the need to develop safer and more effective alternative therapies. This study aimed to investigate the in vitro antitoxoplasmic activity of green-synthesized silver nanoparticles (AgNPs) synthesized using Taxodium distichum extract against tachyzoites of the RH strain of T. gondii.
Materials and methods: Tachyzoites of T. gondii were intraperitoneally inoculated into laboratory mice. After five days, tachyzoites were harvested from the peritoneal cavity and used for subsequent experiments. The antiparasitic activity of green-synthesized AgNPs at concentrations ranging from 50 to 800 µg/mL and that of the methanolic extract of T. distichum at a concentration of 25 mg/mL were evaluated against tachyzoites of the RH strain of T. gondii. Parasite viability was assessed using the MTT assay, while cytotoxicity was evaluated using the J774 macrophage cell line. Data were analyzed using one-way analysis of variance (ANOVA), followed by appropriate post hoc analyses.
Results: AgNPs exhibited significant concentration- and time-dependent antiparasitic activity. Both the AgNPs and the plant extract were significantly more effective than the negative control at all concentrations tested (P < 0.05). AgNPs at concentrations of 400 and 800 µg/mL demonstrated antiparasitic activity comparable to that of the positive control. The IC₅₀ value of green-synthesized AgNPs after 48 h was 41.75 µg/mL.
Conclusion: Green-synthesized AgNPs prepared using T. distichum extract demonstrated promising in vitro activity against T. gondii tachyzoites of the RH strain. These findings suggest that T. distichum-mediated AgNPs could be potential candidates for the development of new therapies for toxoplasmosis. Further in vivo studies are warranted to evaluate their therapeutic efficacy and safety.
