Background and purpose: The potato disc assay is a well-established method for preliminary screening of compounds with potential antitumor activity, based on their ability to inhibit crown gall tumor formation induced by Agrobacterium tumefaciens. The present study aimed to compare the inhibitory effects on crown gall tumor formation of leaf extracts from Zataria multiflora and Taxus baccata with that of the reference drug vincristine.
Materials and methods: In this experimental study, healthy potatoes were surface-sterilized using 1% sodium hypochlorite and cetrimide, then aseptically cut into uniform discs and placed on 1.5% agar medium. A suspension of A. tumefaciens was mixed with varying concentrations of the plant extracts (0.1, 0.25, 0.5, 1, 1.5, 2, and 2.5 mg/mL) and inoculated onto the discs. The discs were incubated at 25 °C for 12–21 days. After incubation, crown gall tumors on each disc were counted, and the percentage of tumor-growth inhibition relative to the negative control was calculated. Statistical analyses were performed to determine differences between groups.
Results: The leaf extract of T. baccata exhibited significant inhibitory activity against crown gall tumor formation at all tested concentrations. In contrast, Z. multiflora extract showed a significant inhibitory effect only at 2 mg/mL, with negligible activity at lower concentrations.
Conclusion: Based on these findings, Taxus baccata extract showed greater inhibitory activity against crown gall tumor formation, whereas the effects of Zataria multiflora extract were limited and concentration-dependent. These results support further investigation using cellular and in vivo models to determine whether these plant extracts have antitumor potential.
